From Buffalo...well, that are a couple of Wyndham's in either Ontario or Quebec that look interesting (they are affiliated resorts, so limited inventory).

New York City--it's a boatload of points! 33K per day for a hotel room, more for a studio or 1BR. There is a hotel type with two double beds instead of one king. They are all 33K per day (more for Friday and Saturday). Parking is horribly expensive, park elsewhere and take the train in. A 7-day Subway pass is $32 per person and you can get to LaGuardia or JFK on the Subway/Express bus.

Old Town Alexandria!! (fewer points, right by the subway/AMTRAK train, free parking!)
National Harbor (more pools, Wi rooms, activities, Kids love it, near the new Casino and Outlet mall, Parking is $$)

Williamsburg--I like how quiet and how few points Patriot Place is. The other resorts look nice.

Atlantic City--Took college kids there for a conference. The resort is nice on the Presidential/Penthouse floors, Atlantic City is kind of seedy. The students went to the board walk and ocean and that was fun for them. It was November, so not a "beachy" feel or very busy.

Chicago--again really a lot of points and parking is ridiculously expensive. Park elsewhere and take the train in.

I would like to go to the resorts in New England but I haven't been there yet.

Nashville is nice and has lots to do. It is not by the down town, but it by the Opryland Mall and Gaylord hotel and conference center...and Opryland.

I agree with the other recommendations in this thread .. lots of wonderful beach, city and destination site resorts in the east.

But I wanted to make a special callout for Carriage Ridge resort in Ontario .. beautiful hilly area about an hour north of Toronto. It backs up to the top of a ski resort .. once walked out the back door for July 1st celebration .. Canada day celebrated much like our July 4th. A small crowd from the resort walked a few steps up to the top of the ski lift to watch fireworks being set off from the valley below. Nice pool / grills .. laid back kind of place. Can drive into Toronto and park at one of their subway parking garages to get a taste of the city life. Really unique décor in the large units too.

Then up in Quebec .. about an hour + north of Montreal in Hippolyte is Geopremier. Built around a little lake. Beautiful area .. smaller units that we've never liked but a nice stay in remote area .. kind of like Maine only you've got the French influence all around you.

We've been to both of these several times and have never had a problem getting in. Will be back again.

Bob
