I would second this strategy/itinerary. I've done it both ways and definitely prefer renting a car one way (walked to Lowe's hotel Avis desk from McAlpin) on the morning we checked out and drove to Key West and returned the car to the airport there. This works perfectly because you absolutely do not need a car in either South Beach or Key West. Then we flew home (via a connecting flight) out of Key West on the last evening of our trip. No back tracking, minimal driving, and no paying for a rental car or parking you don't need during majority of your trip.
Forgot to mention we rented bikes for the duration of our stay in Key West and they were the perfect way to get around. The rental company delivered the bikes to where we were staying and picked them up at the end of our trip. Highly recommend.
