I would choose Harbour Club because it is in SeaPines, which has wonderful bike trails, close to the beach..by bike or car, is in the heart of Harbour Town with shops, restaurants, marina, etc just a few feet away and most of all because we own there and just love it. It is also close to South Beach with all their shops, restaurants, etc and Coligny Circle with the same amenities...shops, restaurants, beach, etc. Even if it's not swimming weather, it is certainly beach weather..
Without question, I'd select Barony. It is in Port Royal Plantation and there are plenty of walking/biking paths. The resort is on the ocean. There is a full service spa and fitness center.
You can always drive to Sea Pines. Harbor Club is recently redone, but it doesn't have the full set of amenities and you have to go to Monarch for some of them (if you care). Of course it is not on the ocean.
Both are very nice and the accommodation are equal. First visit on the Island I would lean toward Harbour Club and Sea Pine Island. The marina and Harbour Town, the famous red & white lighthouse, plus the PGA golf course are what made Hilton Head Island in my opinion.
